| Bu çalışmada, arpa (Hordeum L.) ve fiğ (Vicia L.) bitkilerinin toprak mikroflorasına (toplam aerobik mezofilik canlı bakteri sayısı, aktinomiset sayısı, maya ve küf sayısı) etkisi araştırıldı. Topraktan izole edilen ve bakteriyosin üreten Bacillus cinsi bakteriler klasik yöntemlerle tanımlandı ve moleküler (PZR) yöntem ile de kontrol edildi. Sonuç olarak, arpa ve fiğ ekilen topraklardaki toplam aerobik mezofilik canlı bakteri sayısı, aktinomiset, maya ve küf sayısında genel olarak artış görüldü. İzolatlar içerisinden Bacillus cinsi bakteriler koloni morfolojilerine göre seçildi ve koloni PZR yöntemiyle Bacillus oldukları teşhis edildi. Bacillus izolatları içerisinde B. subtilis, B. mycoides, B. megaterium ve B. amiloliquefaciens’in bakteriyosin ürettiği tespit edildi. Bu 4 tür arasında sadece B. subtilis’te plazmit varlığı belirlendi. | |||
| In this study, effect of Hordeum L. and Vicia L. plants on soil microflora (numbers of total aerobic mesophilic alive bacteria, actinomycetes, yeast and fungus) was investigated. Bacillus species isolated from soil and producing bacteriocin were identified by using conventional methods and these strains were also confirmed via molecular methods using PCR. Our results showed that the numbers of total aerobic mesophilic bacteria, actinomycetes, yeast and fungus on soils growing Hordeum L. and Vicia L. isolated were increased. Among the isolates, Bacillus spp. species were selected according to their colony morphology and confirmed via PCR method. Among the Bacillus isolates, bacteriocin producing of B.subtilis, B. mycoides, B.megaterium and B.amiloliquefaciens bacteria was diagnosed. The only plasmid bearing isolate was B.subtilis. | |||
