| iiiANKARA YÖRES NDE TÜKET ME SUNULANÇ Ğ SÜTLERDE SALMONELLA-SH GELLAVE BAZI PATOJENLER N ZOLASYONU(Yüksek Lisans Tezi)Merve ÜZÜMGAZ ÜN VERS TESFEN B L MLER ENST TÜSÜŞubat 2006ÖZETBu çalışmada 06.10.2003-10.01.2005 tarihleri arasında Ankara ili semtlerindeve ilçelerinde satışa sunulan sokak sütlerinden toplam 100 adet çiğ süt örneğiile çalışılmıştır. Bu örneklerde Salmonella, Shigella, Yersinia, Listeria veStaphylococcus, Citrobacter, Klebsiella, Enterobacter, Providencia, Proteus,Morganella cinsi bakterilerin varlığı araştırılmıştır ve her bir örneğin toplamcanlı mezofil bakteri sayımı, toplam koliform bakteri sayımı ve toplam E. colisayımı yapılmıştır. Ayrıca izole edilen her bir bakterinin proteaz aktiviteleri velipaz aktiviteleri çalışılmıştır. 100 çiğ süt örneğinin toplam mezofil bakteriortalaması 1,6×106 adet/ml , toplam koliform bakteri ortalaması 1,1×106 adet/ml,toplam E. coli sayısı ortalaması 7x 105 adet/ml olarak saptanmıştır. Çalışmadaelde edilen toplam 115 izolatın tür dağılımı şu şekildedir: 1 Shigella (%0,86), 9Salmonella (%7,84), 1 Listeria (%0,86), 32 Staphylococcus (%27,82), 17Citrobacter (%14,8), 13 Klebsiella (%11,30), 17 Enterobacter (%14,8), 3Providencia (%2,60), 16 Proteus (%13,91), 6 Morganella (%5,21). Çalışmada,izole edilen 19 patojen mikroorganizmanın %5’i Shigella sonnei, %26’sı S.typhi, %21’i S.paratyphi, %5’i L.monocytogenes, %43’ü de S. aureus olarakbelirlenmiştir. Araştırmada çiğ sütten izole edilen toplam 19 patojen bakterininCalsium Caseinat Agar’da 37 ºC’de proteolitik aktiviteleri çalışılmıştır. Toplam19 izolattan 13’i (%68,43) proteolitik aktivite gösterirken, 6 (%31,57) izolattaivproteolitik aktivite gözlemlenmemiştir. Thesis)Merve ÜZÜMGAZI UNIVERSITYINSTITUTE OF SCIENCE AND TECHNOLOGYFebruary 2006ABSTRACTIn this study 100 samples of crude milk sold out in the streets at the districts ofAnkara between 06.10.2003-10.01.2005 has been used. The presence of thebacteria Salmonella, Shigella, Yersinia, Listeria and Staphylococcus, Citrobacter,Klebsiella, Enterobacter, Providencia, Proteus, Morganella has been investigatedand the live bacteria count, total coliform bacteria count and total E. coli countfor all samples were realized. The protease activity and lipase activity for all theisolated samples were conducted as well. The total mesophyll bacteria averageof the 100 crude milk samples were 1,6×106 cfu/ml , the total coliform bacteriaaverage were 1,1×106 cfu/ml, total E. coli count average was 7x 105 cfu/ml. Thetype distribution of the 115 isolated in this study is: 1 Shigella (0,86%), 9Salmonella (7,84%), 1 Listeria (0,86%), 32 Staphylococcus (27,82%), 17Citrobacter (14,8%), 13 Klebsiella (11,30%), 17 Enterobacter (14,8%), 3Providencia (2,60%), 16 Proteus (13,91%), 6 Morganella (5,21%). 19 pathogenmicroorganisms were isolated from the 100 crude milk samples. The 19 isolatedpathogen microorganisms were determined as 5% Shigella sonnei, 26% S. typhi,21% S.paratyphi, 5% L.monocytogenes, and 43% as S. aureus. The proteolyticactivities of the 19 pathogen bacteria isolated from crude milk has been studiedat 37 ºC at Calcium Caseinat Agar. 13 (68,43%) of the 19 isolated has shownproteolytic activity while 6 (31,57%) isolated hasn?t shown any proteolyticviactivity.
