Eşme Ayvasının Doku Kültüründe Farklı Karbonhidrat Formlarının Sürgün Çoğalması Üzerine Etkileri

Öz

Türkiye takes the first place in quince production in the world and Eşme cultivar is one of the most grown quince cultivars. Breeding studies need to improve for new quince cultivars. Tissue culture serve as a good tool by shortened the breeding process and offers many conveniences in breeding studies. The success of tissue culture depends on choosing the appropriate material, suitable nutrient medium, and needs to determine the appropriate culture conditions and acclimation environment. In the study presented here, the effects of different carbohydrate types were investigated in the shoot proliferation culture of Quince cultivar ‘Eşme’. Micro shoots of Eşme cultivar were cultured in MS nutrient medium containing 2 mg.l⁻¹ BA+0,25 mg.l⁻¹ GA₃ and 7 g.l⁻¹ agar was added into the medium. One of 30, 50 or 70 g.l⁻¹ of sucrose, fructose, glucose and D-sorbitol were added as carbohydrate sources. While the effects of the sugars and their doses were not made a significant effect on the survival rate of the shoots, shoot proliferation was affected by the carbohydrate type. The shoot proliferation rates increased at 30 g.l⁻¹ dose of each sugar. 30 g.l⁻¹ fructose gave the highest shoot proliferation (4,67 units), followed by 50 g.l⁻¹ sucrose with 3,22 units. Shoot lengths increased at 70 g.l⁻¹ glucose. Callus formation and vitrification were observed during the cultures. Vitrification rates were increased by the increasing doses of sucrose while less vitrification rates were occurred in shoots in environments containing fructose. The results clearly show that different carbohydrates affected micro shoot proliferation rates and quality.

Öz

Ayva üretiminde ilk sıralarda olduğumuz bir meyvedir ve Eşme ayvası yetiştirilen çeşitler arasında öne çıkmaktadır. Ayvada ıslah çalışmaları önemlidir. Doku kültürü ıslah çalışmalarında birçok kolaylığı sunmakta, ıslah süresini kısaltabilmektedir. Doku kültüründe başarı da uygun materyal, besin ortamının seçilmesi, uygun kültür koşullarının ve alıştırma ortamının belirlenmesi ile mümkün olabilmektedir. Bu çalışmada eşme ayvası çeşidinin doku kültürü ile üretiminde çoğaltma aşamasında kullanılan farklı şeker çeşitlerinin etkileri incelenmiştir. Eşme ayvasının in vitro çoğaltılan sürgünleri çoğaltma aşamasında 2 mg.l⁻¹ BA+0,25 mg.l⁻¹ GA₃ içeren MS besin ortamında kültüre alınmış, ortama 7 g.l⁻¹ agar ilave edilmiştir. Şeker kaynağı olarak sakkaroz, fruktoz, glikoz ve D-sorbitolün 30, 50 veya 70 g.l⁻¹ dozları ortama eklenmiştir. Çalışma sonunda sürgünlerin yaşama oranına kullanılan şekerlerin ve dozlarının etkileri önemli bulunmazken, sürgün çoğalması kullanılan şeker kaynağından etkilenmiştir. Sürgünlerin çoğalma oranı şeker çeşitlerinin 30 g.l⁻¹ dozunda yüksek bulunmuştur. En yüksek sürgün çoğalması 30 g.l⁻¹ fruktoz (4,67 adet) meydana gelirken, bunu 3,22 adet ile 50 g.l⁻¹ sakkaroz takip etmiştir. Sürgün boyları ise 70 g.l⁻¹ glikozda daha yüksek bulunmuştur. Kültürlerde kallus oluşumu ve vitrifikasyon görülmüştür. Sakkarozun dozunun artması vitrifikasyonu teşvik etmiş, fruktoz içeren ortamlarda sürgünlerde daha az vitrifikasyon oluşmuştur. Elde edilen sonuçlar farklı şekerlerin sürgün kalitesi üzerine etkili olduğunu ortaya koymaktadır.
